319 S. aureus can induce NLRP1 inflammasome activation in patients with atopic dermatitis

چکیده
Inflammasome activation is needed for the production of IL-1 family cytokines IL-1β and IL-18. Both these have been suggested to influence inflammatory responses in AD, as well genetic association AD with polymorphisms IL18R1 IL18RAP genes has proposed. In this study, we report increased expression inflammasome sensor NACHT, LRR, PYD domains-containing protein 1 (NLRP1) adapter CARD Domain Containing (PYCARD), mRNA levels IL1B IL18 lesional skin patients. Moreover, also found a positive relationship between NLRP1 or caspase-1 activity Interestingly, human primary keratinocytes reconstituted epidermis, tested AD-related induced IL18, while none affected However, when were stimulated same infected pathogen Staphylococcus (S.) aureus, secretion IL-18 was significantly increased, siRNA silencing inhibited addition, colocalization analysis published GWAS eQTL datasets confirmed revealed that two are mainly expressed various types immune cells indicating produced by may affect development AD. conclusion, our data demonstrate colonization S. aureus can cause NLRP1-dependent inflammasome, contribute stimulation inflammasome-related at level. results suggest patients variations pathway be more infection.

Common Allergens in Patients with Atopic Dermatitis
Abstract Background and objective: Being exposed to different allergens, followed by the production of specific IgE, has an important role in causing atopic dermatitis, recognizing the allergens and applying immunotherapy for treatment. We aimed to determine the frequency of common allergens in the patients suffering from atopic dermatitis. Material and Methods: In this descriptive- anal...
متن کاملPrevalence of atopic dermatitis in patients with acne vulgaris
Background and Aim: Atopic dermatitis is a chronic inflammatory disease that is caused by an inflammatory process stimulated by Th2 cells. Acne vulgaris is a chronic inflammatory skin disease. It is associated with an increase in sebum secretion, unusual pilosebaceous keratinization and an increased inflammatory immune response. Propionibacterium acne can induce IL-17 production and Th1/Th17 re...
متن کاملAssociation of LL-37 and IL-31 serum levels with SCORing Atopic Dermatitis (SCORAD) score in atopic dermatitis patients
Background: Atopic dermatitis is a chronic, recurrent inflammation of the skin, accompanied by severe pruritus. Immune system dysregulation and skin barrier defects are associated with the abnormalities in atopic dermatitis. Myriad pieces of evidence have pointed to the major roles of LL-37 and interleukin-31 (IL31)in atopic dermatitis. The studies on atopic dermatitis are still limited i...
متن کاملEvaluation of the Expression of NLRP1 Inflammasome in Patients with Bacterial Septicemia
Background: Septicemia is the most important cause of mortality, especially in hospitalized patients, due to the influence of the immune response by infection. NLRP1 (Nod-like receptor P1) is an intracellular receptor that recognizes microbial-dependent molecular patterns. The main intracellular mechanism of anti-septicemia is still being investigated. The purpose of this study was to evaluate ...
